Desantis was born on September 14, 1978 in Jacksonville to Karen Rogers and Ronald Daniel Desantis. All of his great-grandparents were born in Italy, but his family relocated to America long before Desantis was born.  Desantis attended Our Lady of Lourdes Catholic School and Dunedin High School and joined Yale University after schooling. After Yale, he worked as a professor at Darlington School and later tended Harvard Law School with a Juris Doctor cum laude.

Rob Desantis married Casey Black in 2010. Casey is a former television host for the Golf Channel and WJXT. The couple together have three children. A Roman catholic, DeSantis is a member of the Veterans of Foreign Wars and the American Legion.
